INTRODUCTION. Microplastics (MP) are plastic particles less than 5 mm in size, which can be consumed with water and food. MP impact analysis remains an actual problem.
AIM. To evaluate the ability of MP to induce inflammatory responses in the colon and aggravate the pathologic process in acute colitis in mice.
METHODS. Male C57BL/6 mice were divided into 4 groups 1 control 2 MP exposure 3 acute colitis 4 acute colitis during MP consumption. MP exposure: water was replaced for a suspension of 10 mg/L of polystyrene particles with a diameter of 5 μm (dose 2.3 mg/kg/day). Acute colitis: from day 36 of the experiment, 1% DSS was added for 5 days to the water. Animals were sacrificed on the day 43. The prevalence of ulcers and inflammation, cell infiltration of the intestinal mucosa, reactions of secretory cells, gene expression of glycocalyx components and tight junctions were evaluated.
RESULTS. In healthy mice, MP caused in the mucosa of the distal colon a decrease in the volume fraction of Goblet cells, an increase in the content of highly sulfated mucins in the Goblet cells, an increase in the number of endocrine cells, increased number of cells in the lamina propria, decreased volume fraction of macrophages, increased mRNA expression of Muc1,3, tight contact proteins Cldn2,4,7, proapoptotic factor Bax and anti-apoptotic factor Bcl2. In colitis compared to the control, the volume of Goblet cells in the colon decreased and there was decrease the content of neutral mucins in them, the number of endocrine cells increased, the content of cells in the lamina propria and macrophages was increased; the expression of Cldn4 mRNA increased and there was a tendency to increase Cldn7 and Muc13, the expression of Bcl2 increased. The mRNA level of Bax increased in mice that did not consume MP. In colitis on the during of MP consumption in comparison with colitis without MP there was higher of ulcer-inflammatory process in the colon, decrease in the content of neutral mucins in Goblet cells and increased expression of Muc13 mRNA; the levels of Muc3 and Bax mRNA did not change and the increase in the level of Cldn4 was less pronounced.
CONCLUSIONS. Adaptive reactions of the colon lamina propria were revealed in healthy mice consuming MP. MP leads to a more pronounced course of experimental acute colitis.
